Dimethyl Sulfoxide as a Cryoprotective Agent for Freezing Bovine Semen

Abstract
The effect of DMSO in combination with glycerol on the pre- and post-freeze motility of bovine spermatozoa extended in homogenized, skim milk and egg yolk-Tris extenders was studied. In no case did the addition of DMSO to either milk extender result in a higher mean motility than that obtained with the optimum level of glycerol alone. However, a significant quadratic effect was found in the egg yolk-Tris extender, with 1% DMSO in combination with 6% glycerol providing the highest mean motility. It would thus appear that the addition of DMSO at the levels investigated does not improve the post-freeze motility of bovine spermatozoa extended in either homogenized or skim milk extenders, but it may enhance the motility of bovine spermatozoa extended in egg yolk-Tris extender.
