A Simple Method of Preparation of Gastroinestinal Tract Tissues for Scanning Electron Microscopy

Abstract
Equipment found in many animal science oriented laboratories was used to develop a reliable straight forward procedure for preparing animal soft tissue for scanning electron microscopy. Tissues were fixed in glutaraldehyde and osmium tetroxide, dehydrated in alcohol and dried in a lyophilizer. It was found that freezing was not essential for artifact-free appearance. Charging under the electron beam was further reduced by attaching specimens to SEM mounting stubs with the electrically-conducting cement cited herein. This procedure resulted in gastrointestinal tract surfaces which appeared as previously described by histologists and transmission electron microscopists. Copyright © 1974.
