Rapid Purification of Synthetic Bombesin by Countercurrent Chromatography on the Multi-Layer Coil Planet Centrifuge

Abstract
A temperature-controlled multi-layer coil planet centrifuge rapidly yields highly efficient preparative separations of polar compounds. Capability of the method was demonstrated on a one-step purification of crude synthetic bombesin with a two-phase solvent system composed of n-butanol/dichloroacetic acid/water (100:1:100). Under an elevated temperature at 45.degree. C to 50.degree. C, the bombesin peak was eluted within two hours. Reversed phase HPLC analysis of the bombesin fractions showed over 98% purity.
