In vitro Culture of Western Redcedar (Thuja plicata Donn). I. Plantlet Formation

Abstract
Juvenile and mature tissues of T. plicata were induced to form multiple adventitious buds when cultured in continuous light at 25.degree. C on defined nutrient media containing 1 .times. 10-6 M to 1 .times. 10-5 M BAP [6-benzylaminopurine] in the presence or absence of 1 .times. 10-7 M NAA [naphthaleneacetic acid]. Excision and transfer of adventitious buds onto a wide range of defined and undefined media allowed shoot elongation and leaf formation to occur. Subsequent transfer of the shoots onto media containing 5 .times. 10-6 M to 5 .times. 10-5 M IBA induced adventitious root formation within 4-6 wk. The resulting plantlets exhibited normal shoot morphology and were successfully transferred from tissue culture flasks to greenhouse conditions.
