Restriction Map and Maternal Inheritance of Chloroplast DNA in Eucalyptus nitens

Abstract
A restriction map of the chloroplast genome in Eucalyptus nitens was constructed using five restriction enzymes. Sequential mapping of single and double digests of DNA was carried out using heterologous probes from the petunia and tobacco chloroplast genomes. The chloroplast genome of E. nitens is 151 kb in size and possesses the inverted repeat characteristic of most angiosperms. The genome is essentially colinear with the petunia chloroplast genome and, hence, with angiosperms in general. This similarity enables the use of heterologous probes from petunia to investigate polymorphism in chloroplast DNA of E. nitens. Restriction fragment length polymorphism in the chloroplast DNA of six interpopulation crosses of E. nitens was analyzed using five probe-enzyme combinations. The chloroplast genome was maternally inherited in all progeny of these crosses.
