Abstract
This review describes the design and current development of leukotriene biosynthesis inhibitors as potential antiinflammatory agents. Knowledge of the enzymatic mechanism of 5-lipoxygenase led to specific inhibitors of this enzyme which catalyzes a key step in the leukotriene pathway. Competitive inhibitors include iron chelators, redox agents and alternate substrates. Further, the potential of product inhibitors and mechanism based inactivators of 5-lipoxygenase is discussed.Keywords
