A Cleaning Method for Coccidial Oocysts Using Density-Gradient Sedimentation

Abstract
The density-gradient sedimentation technique has yielded clean Eimeria tenella oocysts. The gradients were produced in an apparatus consisting of 2 Sedgwick-Rafter funnels connected at their bottom, using saturated sucrose solution and distilled water. Twenty ml of oocyst suspension was overlaid with the gradients in a 250 ml centrifuge bottle. A centrifugation for 6 minutes at 1100 g produced layering. The upper layer contained nonviable or empty oocysts, the second pure sporulated oocysts and the third nonsproulated oocysts and host tissue cells while fecal debris was thrown to the bottom.Keywords
