RETARDED ABSORPTION OF PELLETS OF PROTAMINE-ZINC INSULIN

Abstract
K is shown experimentally that protamine-zinc insulin (PZI), obtained as a powder from the commercial batches at pH 7.1, is the only type of insulin useful for subcut. implantation. This PZI powder, compressed in tablet form, alone or mixed with 50% cholesterol, is absorbed so slowly that the lowering of the glycemia of normal rabbits is prolonged for about 50 days. Subcut. or intramusc. implantation of PZI - cholesterol could control alloxanic diabetes of rabbits for approx. 100 days (430-690 U./kg. body wt.). On the basis of the difference of wt. of the pellets before the implantation and after their recovery, it was calculated that 1% of the amt. implanted is absorbed daily. This estimate is appropriate for clinical application, and is recommended for further study in human diabetics.
