An Improved Method for the Purification of Sweet Potato β-Amylase*

Abstract
The method consists of aqueous extraction of the acetone-dried powder, heating (60[degree]C, 10 minutes), precipitation with an equal volume of acetone, ammonium sulfate fractionation between 0.2 and 0.7 saturation, acid treatment (pH 3.6), and crystallization by adding saturated ammonium sulfate solution adjusted to pH 3.6. The re-crystallized preparation possessed about 30-fold specific activity, and was shown to be homogeneous by electrophoretic and ultra centrifugal analyses.
