The Mounting of Bacteria for Electron Microscope Examination

Abstract
Bacteria mounted for electron microscope examination from a dist. water suspension suffer the max. amt. of distortion during the original desiccation. Subsequent introduction into the vacuum of the microscope pro-duces relatively little change. In the case of some organisms which can be grown on solid media much of this distortion can be avoided by the use of a new mounting technique. A collodion sola, is floated over a young colony, allowed to evaporate, removed by floating on a clean water surface, mounting on specimen screen, and shadow-cast. In this technique the colony is removed intact. Flagella and bac-terial excretions are shown in their exact relationship to the bacteria in the growing colony.Keywords
