The potentiometric determination of polypeptides and amino-acids

Abstract
The titration curve of an amino-acid in aqueous sugar soln. may be built up from the curves of the 2 separate components in water. If any reaction occurs between the above substances then the product must possess an acidic dissociation constant almost the same as that of the parent amino acid; hence the potentiometric method cannot be used in the study of this reaction. For any amino acid-alkali buffer, the pH change on adding formaldehyde should be independent of the starting pH, in agreement with the findings of previous workers but at variance with the exptl. results of Frankel and Katchalsky (1937) Biochem. J. 31. 1595.
