Dextran Sulfate as a Displacer for the Displacement Chromatography of Pharmaceutical Proteins

Abstract
Dextran sulfate is successfully employed as a displacer for the displacement chromatography of β-lactoglobulins A and B. Performance characteristics for this separation, such as product recovery vs. product purity and throughput, have been reported. It has been shown that dextran sulfate is desirable as a displacer for pharmaceutical separations with respect to column development, regeneration, and toxicity. The effects of displacer molecular weight on the separation have also been experimentally determined. Based on these results, it has been shown that the relative affinities of the displacers and proteins can be correlated to their characteristic charge, and heterogeneous compounds can be used as displacers as long as all variants have higher affinities than the protein sample. A method for rapidly evaluating the characteristic charge with isocratic elution and the stoichiometric displacement model is presented. Gradient elution chromatography has also been studied as a quick method for determining relative affinities. It has been shown that for the separation being considered, retention volumes correlate well with affinities.
