Anti-staphylococcal activity of a penem, Sch 29482

Abstract
Sch 29482 (SCH), a new broad-spectrum β -lactarn antibiotic, was studied in an in-vitro microtitre system using Mueller-Hinton broth for determining its antistaphylococcal activity. Each of the 68 isolates of staphylococci studied belonged to one of the following six groups, and had the median penem MIC values as noted in parentheses: penicillinase-positive (0.25 mg/l), or -negative (0.25 mg/l) Staphylococcus aureus , methicillin-resistant Staphylococcus aureus (32 mg/l), or coagulase-negative staphylococci (128 mg/l), ‘tolerant’ Staph. aureus (0.12 mg/l), or methicillin-sensitive coagulase-negative staphylococci (≤ 0.12 mg/l). Thus, the penem was highly active against all groups except methicillin-resistant staphylococci. In parallel, the antistaphylococcal activities were determined on the same organisms for seven other β -lactam antibiotics: nafcillin, methicillin, benzylpenicillin, ampicillin, cephalothin, cephalexin and cefamandole. The lowest MIC values against methicillin-resistant strains were noted with cefamandole, but SCH had the lowest MIC values with the other groups of staphylococci.Keywords
