Properties of ATP-dependent H+-transport in rat brain synaptosomes.

Abstract
ラット脳の神経終末形質膜におけるATP依存性のH+輸送をacridine orangeの蛍光減弱により検索した.ATP添加後の膜顆粒内H+蓄積を示す蛍光減弱は,低張処理した神経終末では観察されたが,無処理の神経終末では観察されなかった.低張処理した神経終末のH+輸送活性は反応液のアニオンがCl-かBr-の場合にのみ観察され,Mes-,NO3-,I-およびSCN-では消失した.反応液のK+をCs+,Na+またはLi+に置き換えると,K+>Gs+>Na+>Li+の順にH+輸送活性は減少した.このH+輸送活性は,0.3 mM ethacrynic acidでは対照の39.2%に,10μM 4-acetamide-4′-isothiocyanatostilbene-2,2′-disulfonic acidでは36.3%に,また1 mM 4-aminopyridineでは33.1%にまで阻害されたが,1 mM ouabain,500 μM vanadate, 10 μM picrotoxinおよび100 μMγ-aminobutyric acidでは阻害されなかった.これらの結果から,神経終末において,ATP依存性H+輸送系のATP作用部位は細胞内側表面に存在し,Cl-またはBr-の存在,およびK+のK+チャンネルを介する移動が,H+輸送活性を高めることが示唆された.This publication has 15 references indexed in Scilit:
