Abstract
In this interesting study, Gourh et al. were unable to replicate the association of a connective-tissue growth factor (CTGF) promoter with a polymorphism that was demonstrated by Fonseca et al. in the New England Journal of Medicine {1}. Aside from the usual issues with replication of SNP studies, this disappointing outcome may illustrate the difficulty in using polymorphisms in a disease that is likely to combine a series of etiological events. Rather than dismissing such a finding, the evidence that this polymorphism did have a functional impact in the first study may suggest that one should look for modifying effects on other complex diseases where connective tissue may be important.
