Visualization of Carbonic Anhydrase Activity in Polyacrylamide Gels

Abstract
Carbonic anhydrase activity of insect (cricket) testes homogenates can be demonstrated, after electrophoresis, in polyacrylamide gels buffered at pH 8.1 with Veronal to which a few drops of 0.2% bromthymol blue in 50% ethanol have been added. A distinct yellow band appears within 15-30 sec in the otherwise blue-tinted gel when it is placed in cold CO2 saturated water. Controls consisting of gels containing the separated enzyme gave negative results when placed in cold CO2-free water, CO3-free water buffered at pH 4.5, and CO2-free water at pH 2.9.This publication has 6 references indexed in Scilit:
