Affinity Chromatography of Human Intestinal Alkaline Phosphatase

Abstract
The dye Reactive Yellow 13 when immobilized onto Sepharose 4B is shown to be a highly effective ligand for the affinity chromatography of human intestinal alkaline phosphatase. At least 380-fold enrichment of the enzyme in crude extracts has been obtained in a single step. The dye appears to bind to the active centre of the enzyme, possibly mimicking binding of the HPO^2(4)- ion.Keywords
