A NEW PROCEDURE FOR ASSAY OF BACTERIAL HYDROGENASES

Abstract
A new manometric procedure for assay of hydrogenase activity is described. The method is based on measurement of molecular hydrogen evolved from the reduced form of the "one-electron" dye methyl viologen. A survey was made of cell-free bacterial hydrogenases for activity using the new procedure as well as for activity by methylene blue reduction under an atmosphere of hydrogen. All preparations reputed to contain hydrogenase showed either one or both types of activities. The results of the survey and related experiments are discussed in relation to current concepts of hydrogenase action and it is suggested that microorganisms may contain 2 distinctly different kinds of hydrogenase.Keywords
