Genetic, immunological, and gene dosage studies of mitochondrial and cytosolic MDH variants in maize

Abstract
Genetic analysis has shown that six out of 13 inbred maize lines examined possess only one sMdh gene. When these lines were analyzed using rocket immunoelectrophoresis, it was found that they possessed one-half the amount of sMDH protein as those lines with two sMdh genes. Genetic analysis also has shown that some lines of maize do not possess all four mMdh loci. This is further supported by rocket immunoelectrophoretic analysis showing that lines lacking one or more mMdh genes also have less mMDH protein. In addition, a B-A translocation cross was used to study the effect of one, two, and three doses of the mAdh2 gene on the level of mMDH protein. It was demonstrated that mMdh2 exhibits a gene dosage response. The significance of this finding is discussed.
