Abstract
Mice tolerated two oral doses of sulfa pyridine up to 3 g/kg body weight administered 16–18 h apart or a single intraperitoneal or subcuta neous dose of methadone up to 30 mg/kg body weight. However, combined treatment with sulfa pyridine and methadone at doses stated above signifi cantly increased mortality. -fed mice exhibited increased motor activity, irritability and a Straub reaction a few minutes after methadone administration. This was followed by incoordination, dragging of hind legs and sedation. Death occurred within 5 min to 2 h following methadone administration and was usually preceded by rapid and irregular breathing. The Straub reaction and sedation in mice receiving methadone alone was less pronounced. Equimolar substitution of sulfapyridine by sulfadiazine or sulfathiazole showed similar behaviors and mortality rates.
