Adsorption of Urinary Gonadotrophins on Kaolin

Abstract
A method has been devised for the recovery of urinary gonadotrophins. Its main advantages are a reduction in toxicity of extracts and a marked saving in the amt. of alcohol needed. The method is as follows:[long dash]Adjust the 12- or 24-hr. urine specimen to pH 4.5 with acetic acid. Add 5 vol. % of kaolin (powdered and acid washed in a 20% aqueous suspension), stir thoroughly and then allow to settle. Decant supernatant urine or centrifuge and discard the supernatant. Resuspend the kaolin in water 2-3 times to wash it free of urine. Elute the kaolin with [image] NH4OH in 2 separate portions. Combine the ammonia extracts, add acetic acid to pH 8.5 and centrifuge. Save the clear supernatant, adjust to pH 5.5 with acetic and then add 4-5 volumes of alcohol (95%) slowly while stirring. Set in refrigerator to complete precipitation. Centrifuge and dry the precipitate. Dissolve the dry precipitate in water or phosphate buffer (pH 8) and inject the soln. into immature rats or mice for assay.Keywords
