The efficacy of anthelmintics against third stage larvae ofAncylostoma caninum inMastomys natalensis

Abstract
Investigations have been carried out on the multimammate rat (Mastomys natalensis), orally infected with 1000 third stage larvae ofAncylostoma caninum per animal, to evaluate the larvicidal efficacy of anthelmintics in this paratenic host. The oral or subcutaneous administration of 19 anthelmintics and filaricides revealed good activity for levamisole, cambendazole, and mebendazole against 15-day-oldAncylostoma larvae which were located in the muscular tissue. Similarly, good results were obtained with levamisole and cambendazole against the migrating third stage larvae, by treatment two to six days after infection. The benzimidazole derivatives, thiabendazole, oxibendazole parbendazole, and fenbendazole showed marked activity only at high dosage rates. The anthelmintics methyridine, amoscanate, pyrantel tartrate, morantel tartrate; the microfilaricidal organophosphates dichlorvos, fenthion, and haloxon; as well as the micro-or macrofilaricidal drugs diethylcarbamazine, nitrofurantoin, nifurtimox, suramin sodium, and thiacetarsamide sodium failed to show larvicidal activity even in high dosages. The average larval recovery rate after oral infection with 1000 third stage larvae, in untreated control animals, was 14.85%. In experimentellen Untersuchungen an der “Multimannmate Rat”Mastomys natalensis als paratenischen Wirt wurde nach oraler Infektion mit 1000 dritten Larven vonAncylostoma caninum die larvizide Wirksamkeit verschiedener Anthelminthika untersucht. Die orale oder subkutane Applikation von 19 Anthelminthika bzw. filariziden Verbindungen erbrachte eine gute Wirksamkeit von Levamisol, Cambendazol und Mebendazol gegen 15 Tage alte Larven in der Muskulatur. Bei einer 2 bis 6 Tage alten Infektion konnten mit Levamisol und Cambendazol ähnlich gute Ergebnisse erzielt werden. Die Benzimidazol-Derivate Thiabendazol, Oxibendazol, Parbendazol und Fenbendazol zeigten nur nach Verabreichung hoher Dosen eine deutliche Wirksamkeit. Die Anthelminthika Methyridin, Amoscanat, Pyrantel-tartrat, und Morantel-tartrat sowie die mikrofilariziden Organophosphorverbindungen Dichlorvos, Fenthion und Haloxon zeigten, ebenso wie die mikro-oder makrofilariziden Präparate Diethylcarbamazin, Nitrofurantoin, Nifurtimox, Suramin, und Thiacetarsamid auch in hohen Dosen keine nennenswerte larvizide Wirksamkeit. Als durchschnittliche Ansitzrate konnte nach der oralen Infektion von jeweils 1000 dritten Larven pro Tier bei den unbehandelten Kontrolltieren 14,85% ermittelt werden.Keywords
