The enzymic activation of cholic acid by guinea-pig-liver microsomes

Abstract
The synthesis of cholylhydroxamic acid by liver preparations was demonstrated. This was found to be due to 2 distinct mechanisms. The 1st is a simple condensation of cholic acid and hydroxylamine, and is probably lipase-cata-lyzed. The 2nd occurs in the microsome fraction of guinea pig liver and requires the presence of CoA, adenosine triphosphate (ATP), cysteine and Mg+ or Mn2+ for full activity. The results suggest that the following reactions take place in liver micro-somes: (1) cholic acid +ATP + CoA- cholyl - CoA. (2) cholyl-CoA + hydroxylamine[long dash]cholylhydroxamic acid +CoA.Keywords
