Abstract
We previously described the use of an established reverse genetics system for the generation of recombinant human influenza A viruses from cloned cDNAs. Here, we have assembled a set of plasmids to allow recovery of the avian H5N1 influenza virus A/Turkey/England/50-92/91 entirely from cDNA. This system enables us to introduce mutations or truncations into the cDNAs to create mutant viruses altered specifically in a chosen gene. These mutant viruses can then be used in future pathogenesis studies in chickens and in studies to understand the host range restrictions of avian influenza viruses in humans. Nota de Investigación—Desarrollo de un sistema de genética reversa que permite el rescate del virus recombinante de influenza aviar A/Turkey/England/50-92/91 (H5N1). Nosotros, describimos previamente la utilización de un sistema establecido de genética reversa para generar virus recombinantes de influenza A humana a partir de moléculas clonadas de ADN complementario. En el presente trabajo hemos armado un conjunto de plásmidos que permiten la recuperación del virus H5N1 de influenza aviar A/Turkey/England/50-92/91 utilizando ADN complementario. Este sistema nos permite introducir mutaciones o deleciones en el ADN complementario para crear virus mutantes modificados específicamente en un gen seleccionado. Estos virus mutantes pueden entonces ser utilizados en futuros estudios de patogénesis en aves y en estudios para comprender las restricciones de huésped de los virus de influenza aviar en humanos.
