HYDROLYSIS OF URINARY NEUTRAL 17-KETOSTEROID CONJUGATES I. COMPARISON OF VARIOUS PROCEDURES

Abstract
Five procedures for hydrolyzing urines separately and then extracting 17-keto-steroids (17-KS), and 2 for simultaneous hydrolysis and extraction were compared to determine their relative yields of neutral 17-KS and dehydroisoandrosterone-reacting (D-reacting) materials. Max. yields were obtained by 2 methods: (1) by acidifing with 0.1 vol. 70% (v/v) H2SO4; heating in boiling water for 30 min., cooling and extracting with ether; (2) acidifying with 0.1 vol. 40% (v/v) H2SO4, adding benzene, and refluxing for 30 min. Even these methods destroy at least part of the D-reacting substances.Keywords
