Alterations in cardiac oxygen consumption under chronic pressure overload

Abstract
Hypertension and resulting left ventricular hypertrophy was induced in young male Wistar rats (60 to 70 days old) by narrowing of one renal artery (Goldblatt II). 8 and 24 weeks after operation, myocrdial oxygen consumption was measured on a modified in situ heart-lung preparation with nearly isovolumetric left ventricular contractions. Measured myocardial oxygen consumption was related to left ventricular wall stress. The myosin isoenzyme pattern of each heart was determined with pyrophosphate gel electrophoresis. Oxygen consumption related to wall stress averaged over the entire heart cycle amounted to 15 μmoles O2/g·min 8 weeks after operation, and 24.4 μmoles O2/g·min in age-matched controls (Δ 38%, p2/g·beat, and 0.149 μmoles O2/g·beat in the controls (Δ 25%, p2/g·min, in age-matched controls 20.5 μmoles O2/g·min (Δ 21%, p2/g·beat, and in the controls 0.107 μmoles O2/g·beat (Δ 25%, p2, 15 μmol O2/g·min, bei den gleichaltrigen Kontrollen 24,4 μmol O2/g·min (Δ 38%, p2 war der Sauerstoffverbrauch der hypertrophierten Herzen 0,112 μmol O2/g·Schlag, während die Kontrolltiere 0,149 μmol O2/g·Schlag verbrauchten (Δ 25%, p2/g·min, die gleichaltrigen Kontrollen 20,5 μmol O2/g·min (Δ 21%, p2/g·Schlag, die Kontrollen 0,107 μmol O2/g·Schlag (Δ 25%, p<0,005). Der prozentuale Anteil von VM-3 war von 33,5% bei den Kontrollen auf 43,2% bei den hypertrophierten Herzen angestiegen (Δ 29%, p<0,05). Nach den vorliegenden Ergebnissen beruht der verminderte Sauerstoffverbrauch des druckhypertrophierten Herzens auf einer Umverteilung der Isoenzyme von Myosin. Die durch die Zunahme von VM-3 bedingte Transformation des Myokards in einen langsameren, aber ökonomischer arbeitenden Muskel wird als Adaptationsprozeß gedeutet.This publication has 30 references indexed in Scilit:
