Effect of Ectopic Pituitary Homograft on the Physiological Status of Rat Testis

Abstract
Summary: Pituitary glands grafted under the kidney capsule provide a continuous supply of prolactin due to the absence of hypothalamic inhibitory factor. Increase in the level of prolactin in the rat may affect availability of hormone for reproductive function. Single pituitaries were transplanted under the kidney capsule of male rats. After six weeks of the transplant, the recipient rats were sacrificed and trophic activity of testosterone and intratesticular functions were assessed. The weights of ventral prostate and seminal vesicles, full and empty, showed significant increase from the control value. The activity of β‐glucuronidase in kidney, an androgen inducible enzyme, also increased in comparison with the control value. Incorporation of32P in the nucleic acid fraction showed an increase. The increased rate of testosterone production and an alteration in spermatogene‐sis was associated with increased activity of acid phosphatase and decreased activity of β‐glucuronidase. The active mediation of lysosomal enzyme in testicular function as altered by prolactin was indicated. Zusammenfassung: Unter die Nierenkapsel transplantierte Hypophysen verhindern ei‐ne kontinuierliche Prolaktinversorgung durch das Fehlen des hypothalamischen inhibito‐rischen Faktors. Ein Prolaktinanstieg kann die Hormonwirkung auf die Fortpflanzungsfä‐higkeit bei der Ratte beeinflussen. Einzelne Hypophysen wurden bei männlichen Ratten unter die Nierenkapsel transplantiert. 6 Wochen nach der Transplantation wurden die Empfängerratten getötet und Testosteronaktivität wie intratestikuläre Funktionen beur‐teilt. Das Gewicht der Prostata und der Vesicula seminalis zeigten im gefüllten wie im lee‐ren Zustand einen signifikanten Unterschied zu den Kontrollwerten. Die Aktivität des An‐drogen‐induzierten Enzyms β‐Glucoronidase in der Niere stieg ebenfalls gegenüber den Kontrollwerten an. Inkorporation von 32Phosphat zeigte einen Anstieg der Nukleinsäuren‐fraktion. Der Anstieg der Testosteronproduktion mit Veränderung der Spermatogenese war assoziiert mit gesteigerter Phosphatase‐ und erniedrigter β‐Glucoronidaseaktivität. Die aktive Beeinflussung der lysosomalen Enzyme auf die Hodenfunktion weist auf Verände‐rungen durch Prolaktin hin.Keywords
