Enumeration of Bovine Spermatozoa by Packed Cell Method

Abstract
A total of 156 semen samples from 12 Holstein-Friesian bulls were observed and packed cell volume, volume of ejaculate, motility, percent live cells, percent abnormal cells and cell concentration were determined. These suggest that volume of ejaculate, percent motility, percent live cells and percent abnormal cells have no effect on packed cell volume. Concentration as determined by a method utilizing a photoelectric colorimeter was highly correlated (r = .91) to packed cell volume. The mean packed cell volume was 6.3% with a range of 2.0 - 21.0%. These data suggest that packed cell volume is sufficiently accurate to merit its use in determining cell concentration.Keywords
