Purification of the spherule-endospore phase ofCoccidioides immitis

Abstract
Upon shaking an aqueous suspension of spherules, endospores and mycelium of Coccidioides immitis with chloroform, selective agglomeration of the mycelium around the emulsified chloroform droplets occurred. The mycelial strands became aligned on the surface of the droplets in parallel configurations, as an apparent consequence of surface forces, and the droplets accumulated at the chloroform-water interface. Spherules and endospores were not so-affected and could be resuspended readily in the aqueous phase of the system. This observation formed the basis of a procedure that was employed to obtain the spherule-endospore phase from cultures of C. immitis in highly purified form. Les bactéries et les champignons de la couche cornée peuvent être facilement colorés dans les prélèvements effectués par la technique du “scotch-tape” (rubans n° 681 et 473). Parmi les diverses méthodes de coloration qu'il est possible d'utiliser, on emploie les techniques de Hotchkiss-Mac Manus, de Gram et de Giemsa, en lumière normale et, en lumière ultra-violette, l'orangé d'acridine. D'autres applications de cette méthode, comme par exemple l'examen direct des cheveux parasités, les cultures à partir d'échantillons prélevés de la couche cornée, ainsi qu'une méthode rapide de coloration pour le diagnostic sont décrites.Keywords
