Prothrombin Utilization during Clotting: Comparison of Results with the Two-Stage and One-Stage Methods.

Abstract
Changes in prothrombin during and after clotting were followed in normal human and canine bloods in glass and silicon tubes, in platelet-poor human plasma and in canine hemophilic blood. Early in clotting, divergent results were obtained by the 2 prothrombin methods, with a hypoactive and then a hyperactive phase of varying duration observed by the one-stage test. The hyperactive phase varied from 30 min. in dogs to 7 hrs. in platelet-poor expts. Following the hyperactive phase, both methods showed a steady decline in prothrombin, but with one-stage values always at higher levels.Keywords
