Inhibition of Prothrombin Activation in Sodium Citrate Solutions.

Abstract
The autocatalytic activation of prothrombin to thrombin, accelerated by 3-chloro-4.4''-diaminodiphenyl sulfone or by thrombin is inhibited by plasma and serum. A protein solution, obtained from serum by BaCO3 or BaSO4 adsorption and elution with sodium citrate and fractionated with ammonium sulfate, also inhibits the activation when used at a concentration of 2.5 mg/ml. The rate of activation is slower and final yield less. Plasma eluates prepared in a similar manner behaved likewise. The presence of this inhibitor in certain Factor VII or prothrombin concentrates obtained by a similar method may prevent its conversion to thrombin in the presence of 25% Na citrate.
