The Proteolytic Activity of Gastric Juice

Abstract
It was not possible to confirm the hypothesis of Freudenberg and Buchs concerning the existence of a gastric cathepsin. It is stressed that the structure of the protein substrate is the rate-determining factor for peptic digestion in the weakly acid region. The more extensively the substrate molecule is denatured (i.e., the more its peptide chains are expanded) the more readily will it be hydrolyzed in this pH range. In no case (even with 5 different substrates studied by 3 different methods) was any indication found of a 2-peak pH vs. activity curve for proteolytic breakdown by pepsin or by gastric juice.
