Isolation and Characterization of a Novel Disialoganglioside Containing N-Glycolylneuraminic Acid from Pig Skeletal Muscle

Abstract
A novel disialoganglioside containing N-glycolylneuraminic acid was isolated from pig skeletal muscle by DEAE-Sephadex A-25 and latrobeads column chromatog raphy. The content of this ganglioside was 0.8 nmol of total lipid-bound sialic acid/g fresh tissue, which accounted for 12% of the disialoganglioside fraction. On the basis of carbohydrate analysis, permethylation, periodate-oxidation followed by reduction, and sialidase degradation studies, the structure of this ganglioside is characterized to be: Gal(β13)GalNAc(β14)Gal(β14)Glc(β11)ceramide2αNeuGc2αNeuAc
