Modification of the Radiation Effect on the Reproductive Capacity of Tumor Cells in Vivo with Pharmacological Agents

Abstract
The reproductive capacity of tumor cells from the ascites P-388 leukemia of the DBA/2 strain mouse was studied after treatment with 5-Iododeoxyuridine (IUdR) or 5-Bromodeoxyuridine (BUdR) and/or X-irradiation Experimental criteria werethe numbers of leukemic cells from control and variously treated tumor-bearing donor animals necessary to produce 50% takes in a group of recipient mice of the same strain. At doses of 300 mg/kg/day for 4 days, IUdR alone did not affect the reproductive capacity of leukemic cells but markedly enhanced radiation effects upon them (i.e., decreased the number of cells reproductively intact after given single X-ray doses). The amount of radiation potentiation was proportional to the dose of IUdR used. BUdR alone produced a moderate inhibition of tumor cell reproductive capacity but also potentiated radiation effect at low drug dose levels.
