THE MEASUREMENT OF OESTROGENS IN THE URINE OF WOMEN, WITH ESPECIAL REFERENCE TO HYDROLYTIC PROCEDURES

Abstract
The authors compared the results from various hydrolytic meaures applied to aliquots of a single sample. Optimum conditions for the hydrolytic cleavage of the conjugates of estradiol, estrone and estriol, as they are normally excreted in the last trimester of pregnancy, were investigated on 12 urine specimens from 3 women. Comparative results, using Bioassay isotope studies and the brown method of chemical assay led to two-step procedures, (Described) each of which gave maximum recovery for its particular system. The effect of refluxing each urine with zinc and acid (Tzn) upon the yields of estradiol, estrone and estriol was investigated and, although estrone is converted in part to estradiol, conversion of some other urinary constituent was responsible for the major portion of the estradiol found in Tzn hydrolized aliquots. Estriol was not affected. Preliminary Bioassay findings indicated that estrogenic compounds other than estradiol, estrone, and estriol were produced.
