Abstract
Quinol, resorcinol, phenol, m- and p-cresol, pyrocatechol were found to depress the anti-thrombic activity of human serum, with pyrocatechol most effective. This was adapted to the 2-stage assay of prothrombin. Test plasma (0.3 ml) is added to 0.15 ml thromboplastin, 0.6 ml 0.025 M. CaCl2, 0.45 ml 0.15 M NaCl, 0.3 ml 0.18M pyrocatechol in Veronal buffer pH 7.4. After 1-2 minutes 0.2 ml is blown into 0.2 ml human-Ba plasma + 0.05 ml 0.18 M pyrocatechol and the clotting times determined. Successive samples are taken until the clotting time is identical on 3 successive samples.
