PREPARATIVE ISOLATION AND PURIFICATION OF PHENYLETHANOID GLYCOSIDES FROM THE EXTRACT OF FAECES OF BEAGLE DOGS BY HIGH-SPEED COUNTERCURRENT CHROMATOGRAPHY

Abstract
High-speed countercurrent chromatography (HSCCC) was applied to the separation of acteoside and isoacteoside from a crude extract from canine faeces after the dogs were orally administered phenylethanoid glycosides (PhGs). Two-step separation with a quaternary two-phase solvent system composed of ethyl acetate-n-butanol-ethanol-water (system 1, 35:6:6:50, v/v; system 2, 30:10:6:50, v/v) was used. HPLC analysis of final CCC fractions revealed that the two main PhGs showed purity of over 95%. Their chemical structures were identified by 1HNMR, 13CNMR, FAB-MS.Keywords
