Effect of Reserpine on Peptic Ulceration and Gastric Blood Flow in Dogs.

Abstract
Rauwolfia alkaloids have found wide acceptance in the treatment of various anxiety states and hypertension. However, one of the complicating conditions found in the use of this drug has been peptic ulceration with perforation or hemorrhage. To evaluate the mechanisms involved in the formation of peptic ulcers with rauwolfia alkaloids cats and dogs were given histamine in beeswax alone, histamine in beeswax plus reserpine and reserpine alone to see whether reserpine would abet peptic ulceration. In the second portion of the study the effect of reserpine on gastric blood flow was studied by measuring the venous outflow. Reserpine enhanced histamine-induced peptic ulceration from a control level of 44% to 100% in dogs and from 62% to 89% in cats. In addition, the average time of ulceration was greatly shortened. Furthermore, reserpine alone was a veryp otentulcerogenic agent. When 2.5 mg of reserpine was given to dogs and the blood flow measured by venous outflow method a significant decrease occurred. It is concluded that reserpine alone and in combination with histamine is an ulcerogenic agent. One of the mechanisms involved may be decreased blood flow to the stomach following its administration.Keywords
