Disc-gel electrophoresis of + and - strains and of gymnothecial cultures ofArthroderma tuberculatum

Abstract
Disc-gel electrophoresis of 26-day-old petri plate cultures of 3+ and 3- strains of A. tubenulatum, grown separately and together (3181 × 3182 and 1593 × 1967) to form gymnothecia on modified cereal agar, was performed for their protein, peroxidase, esterase, and polyphenol oxidase patterns. There were more protein and peroxidase bands in + strains that in the — strains. When crossed, gymnothecial cultures produced 1·5 to 2 times more protein bands than when grown separately, but the peroxidase isozymbes were the same in number in crossed cultures as in — strains. More esterase bands were found in — strains than in + strains and in the crossed cultures. The number of esterase isozymes in + strains and in the gymnothecial cultures was the same, but their patterns appeared to be somewhat different. None of the + and - strains nor the gymnothecial cultures showed polyphenol oxidase activity. The results indicate that the mating types of A. tuberculatum differ from each other in their protein, peroxidase, and esterase isozymic patterns. Getreideagar-Kulturen von je drei getrennt gewachsenen und zur Bildung von Gymnothecien gekreuzten (+)- und (-)- Stämmen von Arthroderma tuberculatum wurden mittels Disk-Elektrophorese auf Eiweisse, Peroxydasen, Esterasen und Polyphenoloxydasen gepruft. Die Eiweiss- und die Peroxydase-Banden waren bei den (+)- Stämmen breiter als bei den (-)- Stämmen. Die Peroxydase-Banden der gekreuzten Kulturen waren gleich wie diejenigen der (-)- Stämme, doch zeigten die gekreuzten Kulturen 1,5 bis 2mal mehr Eiweiss-Banden als die getrennt gewachsenen. Bei den (-)- Stämmen wurden mehr Esterase-Banden gefunden als bei den (+)- Stämmen und den gekreuzten Kulturen. Die Anzahl der Esterasen war bei den (+)- Stämmen gleich wie bei den gekreuzten Kulturen, doch stimmte die Lage der Banden nicht genau überein. Weder bei getrennt gewachsenen noch bei gekreuzten Kulturen fand sich eine Polyphenoloxydase-Aktivität. Somit bestehen zwischen den beiden Paarungstypen von A. tuberculatum gewisse Unterschiede im Muster der Proteine, Peroxydasen und Esterasen.Keywords
