Studies with the Agar Cup-Plate Method

Abstract
The following factors may influence the results obtained with the agar cup-plate method of testing antiseptics: strain and species of organism, pH of the medium, agar conc., size of the bacterial inoculum, thickness of the agar medium, and temp. of incubation. A standardized technique was developed. This technique is comparatively simple and yields consistent results with 3 representative antiseptics: phenol, HgCl2, and crystal violet. The recommended method is described in detail.
