Abstract
A new method called FPi analysis was previously used to analyze time sequences of DNA histograms taken from a perturbed population of cells. The method can be used to analyze the in vivo cell cycle kinetic response of bone marrow and of lung metastases of the B16 melanoma tumor in mice to chemotherapeutic agent. The technique allows useful kinetic data to be obtained with minimal processing of the raw histograms, allowing fast analysis of the data. To monitor the kinetic response of living tissues, it is essential to collect multiparameter distributions; to monitor only the 1-dimensional fluorescence histogram can give misleading results. Using these multiparameter histograms, the growth fraction of the lung metastases during treatment can be monitored, allowing discrimination between cell synchrony and cell recruitment from the resting compartment.Keywords
