CHEMICALLY DEFINED MEDIUM FOR THE GROWTH OF PASTEURELLA TULARENSIS

Abstract
A chemically defined medium containing 13 amino acids, KH2PO4, NaCl, MgSCO4, FeSC4, spermidine phosphate, thiamine and glucose was formulated which supported the growth of several virulent strains of Pasteurella tularensis in viable cell yields comparable to those obtained in complex media (18 to 55 billion cells per ml) after 16 to 24 hr. incubation at 37[degree] C. The medium permitted the initiation of growth of the virulent Schu S-5 strain from an inoculum of approximately 10 cells per ml. Growth of this strain from larger inocula was supported through 5 serial transfers without reduction in viable cell yields. Strains of reduced virulence required the addition of calcium pantothenate or uracil, adenine, and guanine for maximal viable cell yields of 0.7 to 13 billion per ml.Keywords
