Sphingosine as Inhibitor of Prothrombin Activation With Thromboplastin or With Threone

Abstract
The activation of purified prothrombin with calcium ions, Ac-globulin and lung extract thromboplastin is inhibited with sphingosine prepared from natural sources and also with synthetic dl-sphingosine. Likewise, the conversion of purified prothrombin to thrombin with calcium ions, Ac-globulin and threone is inhibited; however, in low concentration sphingosine tends to accelerate thrombin formation. By definition threone consists of platelet factor 3 and platelet cofactor I. Either of these two substances, as purified concentrates, can overcome the inhibitory effect of sphingosine when added in large amounts.
