ANTIGENIC AND RELATED BIOCHEMICAL PROPERTIES OFLISTERIA MONOCYTOGENESI

Abstract
By sonic oscillation and differential centrifugation, 30 to 50% of the total cell wall material of L. monocytogenes was recovered. The washed cell wall contained about 20% hexose represented by equal concentrations of glucose and galactose, and about 5% hexosamine. Although about 50% of the wall material was positive as Kjeldahl protein, only the following amino acids, at relative concentrations indicated, were present: alanine, ([image]), glutamic acid ([image]), ([alpha], [epsilon] ) diaminopimelic ([image]), aspartic ([image]), and leucine ([image]). The remaining 25% of the wall was not accounted for quantitatively. However, the washed wall material contained at least 5% of the P, 15% of the Fe, and 36% of the Ca content of the entire cell, and may contain as much as twice these values since only 30-50% of wall material was recovered. Kinetic plots of release of Fe and P during cell breakage showed that Fe originates in the wall fraction and P in both wall and protoplasm. Results with Fe suggested that the wall fraction contains adhering cytoplasmic membrane material. UV spectra of wall material failed to reveal presence of nucleic acid. The cell wall or membrane may be the site of the monocyte producing factor.Keywords
