Purification and Isolation of Certain Intermediates Formed Prior to the Activation of Prothrombin

Abstract
Using a new centrifugation technique, it was possible to isolate and purify 2 intermediate reaction products which are formed in the early stages of the normal clotting of blood. One of these products is formed through the interaction of thromboplastin and labile factor (accelerator globulin or accelerln). It was isolated by centrifugation. The other product is formed by reacting thromboplastin, stable factor (co-thromboplastin, SPCA, or convertin), and Ca++. It was also isolated by similar techniques. Normally, both reactions proceed side by side, and finally a complex intermediate is formed which can also be isolated and purified by centrifugation. In this completed form, it can react with Ca and purified prothrombin to form thrombin.Keywords
