Experimental Tubular Nephritis Produced by Safranin O

Abstract
The intraven. admn. of the dye Safranin O to rabbits and dogs results in selective renal injury, consisting of epithelial necrosis essentially limited to the proximal convoluted tubules. Minimal injury which is rapidly and completely repaired follows an intraven. dose of 10 mg./kg. in rabbits. Admn. of 20-40 mg./kg. results in moderate to practically entire necrosis of the renal proximal convoluted tubules. The histologic picture is essentially identical with that produced by the heavy metals, particularly bichloride of mercury. Evidence is cited for the suggestion that this nephrotoxic action of safranin, as well as the essentially identical action of certain other organic nephrotoxic compounds, parallels the action of the heavy metals biochemically as well as pathologically, and that these organic compounds exert their nephrotoxic effect by acting as inhibitors of vital enzyme systems.Keywords
