Abstract
Crosses were made between streptomycin sensitive (ss), resistant (sr), and dependent (sd) forms of the bacterium E. coli (using mutants of the lines developed by Dr. J. Lederberg from the sexually fertile strain K 12). Resistance and dependence were inherited as if controlled by allelic forms of the same gene locus. This locus is linked with the genetic marker loci controlling fermentation of maltose, xylose, D-galactose, and L-arabinose, but not in a manner which would indicate an unambiguous linear sequence of the genes concerned. No linkage was observed with the factors for lactose fermentation, phage T1 resistance, and thiamin requirement. Four double mutants which had reverted from streptomycin dependence to independence (two to sensitivity, and two to resistance) were also crossed with the ss and sr forms. Three of these behaved as if the 2d of the two mutations had taken place at the same locus as the first; one (a resistant) appeared to be the result of a 2d mutation involving a modifier locus. It is suggested that there is in all probability a very extensive series of multiple alleles affecting streptomycin response.
