Connective Tissue VI: Synthesis of Collagen by Rat Uterine Slices.

Abstract
An in vitro system for collagen biosynthesis has been devised by the use of rat uterine slices. Tissue slices were incubated in Krebs-Henseleit medium with uniformly labelled C14 -lysine, at pH 7.35 - 7.45, under 95% O2 and 5% CO2 atmosphere, at 37[degree]C in a Dubnoff incubator. The presence of Cl4-nydroxylysine was accepted as a measure of collagen synthesis. The radioactivities of isolated collagen, hydroxylysine and lysine were increased by the increment of the amount of C14-lysine in the incubation medium. The collagen synthesis was increased with increase of incubation time up to 6 hours. The capacity for synthesizing collagen in uterine slices was an inverse function of the age of the donor animal. It is believed results can be standardized sufficiently well for the application of this method to bioassay work.Keywords
