Endocrine Pharmacology of Methyl Reserpate Derivatives

Abstract
Five derivatives of methyl reserpate were compared with reserpine in rats concerning the effects on growth, organ weights, estrus, fertility, ACTH release, blood pressure in adrenal regeneration hypertension, and on electrolyte and water excretion. The relative potency of the new compounds differed markedly both among themselves and from reserpine, depending on the parameters measured. Tetrahydropyranyl methyl reserpate (Su-7064) showed the greatest specificity in suppressing estrus and fertility, in stimulating ACTH release and in altering kidney function. In these respects its activities equaled or exceeded those of other compounds which had greater sedative, hypotensive and growth-retarding potencies. Under the conditions used, the ACTH-releasing effect of reserpine was blocked by iproniazid, whereas that following exposure to cold was only partially blocked.Keywords
