COMPARATIVE STUDY OF THE 17-KETOGENIC (NORYMBERSKI), GLENN-NELSON AND REDDY METHODS FOR THE DETERMINATION OF C21URINARY STEROIDS

Abstract
A series of urine samples was analyzed for 17-OH-corticosteroids by the 17-ketogenic steroid method of Norymberski, the Glenn-Nelson method and the Reddy technique. The results indicate relatively good correlation between the Norymberski and Glenn-Nelson values and the clinical findings, but serious discrepancies were obtained with the Reddy method, especially in urines containing only small amounts of corticosteroids. Utilizing 3 wavelengths for reading with the latter procedure, although frequently altering the values, did not necessarily improve their agreement with those obtained by the other 2 methods nor with the final diagnosis. Data are presented which indicate that the Glenn-Nelson technique leaves a significant percentage of the 17-OH-corticosteroids in the residual urine following hydrolysis and extraction. The results point up the advantage of the Norymberski procedure in the diagnosis of the adrenogenital syndrome, a condition in which pregnanetriol is excreted in abnormal amounts and measured as a 17-ketogenic steroid.Keywords
